How can targeted keywords index in the search engine? Do I need to promote the site using that keywords?

you can use your keyword as sig in some forums... or a title as your link..

leftybogs was right, use your keyword as anchor text in your signature from forums or posting to groups,sig in your articles, blog, classified ads and when doing link exchange.

make sure that your content is relevant on your targeted keywords, learn the basic of SEO first, then back to this forums for your questions. you must know first the basic so we can talk here and we help much better.
